   Ik zit al jaren in uniqure en wacht een overname af. Met tigenix en ablynx heeft mij dat geen windeieren gelegd terwijl ik spijt heb van premature verkoop Galapagos.

   Ik ben ook vanaf A.M.T. belegger rechtstreeks en via Gilde Health Care voor een vrij grote positie aandeelhouder en heb de gehele neergang en vervolgens de
   opgang van het aandeel meegemaakt.
   
   Ik denk dat ik ook maar blijf zitten.
   In afgelopen tijd diverse keren hoog verkocht en laag bijgekocht, heeft per
   saldo een goed rendement opgeleverd.

   Iedereen gefeliciteerd die in UniQure is blijven zitten. Zelf ben ik twee jaar geleden uitgestapt omdat ik de dalingen op een gegeven moment niet meer kon aanzien. Ik dacht mijn verlies toen te kunnen beperken. Had ik 2 jaar geduld gehad dan stond ik nu zo ongeveer weer op het BreakEvenPoint. Ach ja, beleggen blijft lastig.

    Cumulative Preclinical HD Data Provides Optimism Ahead of IND in 2H18
    • Bottom Line: Antisense oligonucleotides (ASOs) appear to be effective in targeting and suppressing mutant huntingtin (mHTT) protein in Huntington's disease (HD) pts. A recent post hoc analysis shows a correlation between mHTT suppression and functional improvements (see our colleague's take HERE) raising the hopes of a disease-modifying HD drug. Unlike repeat administrations required of ASOs (intrathecal); however, uniQure has a gene therapy candidate that could deliver durable suppression of mHTT with a single administration. uniQure's preclinical data presented at the American Academy of Neurology (AAN) is encouraging, in our view,
    ahead of IND filing in 2H18. Reiterate OP on QURE.
    • Extensive preclinical data warrants advancement of QURE's HD gene therapy candidate. uniQure's data encompasses both in vitro (induced pluripotent stem cell/iPSC) and in vivo (rodent, primates, and minipig) models all demonstrating effective transduction and consequent suppression of mHTT mRNA and protein. Effects were durable (12 mos. in mice; 9 mos. in minipig) and appear to facilitate improved functionality in mice (as measured by clasping and rotarod tests) and also a greater survival benefit in mice. To this last point, treated mice (eg. R6/2 model) showed a median improvement of 29 days (vs. vehicletreated; p=0.0292). With the usual caveats required for performing crosstrial comparisons being underscored since the data is preclinical, we find it interesting that this corresponds fairly well with the survival benefit demonstrated by Ionis and Roche's ASO IONS-HTTRx/RG6042 in the same animal model.
    • Gene therapy may target mHTT where it matters; 40-50% protein reduction could be therapeutic. Based on in vitro and in vivo studies, uniQure has noted prevention of aggregate formation and pathology changes when mHTT protein levels are reduced by 40-50%. Although Ionis (IONS [NR]) has also achieved 40% mHTT protein reduction on average in their Ph.1/2 study, uniQure believes its intrathecal administration could leave much of the relevant parts of the brain unaddressed. In contrast, by leveraging AAV5's tropism and intrastriatal injection, gene therapy can deliver the cargo in the caudate and the putamen, 2 structures where the disease starts in HD pts. And
    beyond these structures, evidence suggests AAV5 can jump synapses to deliver its cargo elsewhere in the brain as the disease advances further broadening the therapeutic effect.
    • Long-term ramifications of suppressing wildtype HTT protein remain TBD. In the midst of the promising preclinical data, one uncertainty has to do with suppressing wildtype HTT. Whereas Wave(WVE [OP]) has adopted a more specific approach targeting particular single nucleotide polymorphisms (SNP) of mHTT in HD, it appears uniQure may suppress both wildtype and mutant HTT. uniQure has conducted toxicity experiments in multiple models, with a Good Laboratory Practice (GLP) tox study ongoing in primates, that have yet to culminate in noteworthy toxicity events.
    • IND filing expected in 2H18. Though early, we're inclined to believe positive preclinical data across multiple animal models could potentially materialize into meaningful clinical benefit. With the caveat of cross-study
    comparisons, uniQure's survival benefit in mice also appear better than IONIS-HTTRx. uniQure has guided to an IND filing (and clearance) in 2H followed by an expeditious entry into the clinic.

    Als je alles in google translate knipt en plakt is het resultaat soms wat houterig maar 95% correct. Dit is bijvoorbeeld de google translate vertaling van de 2de alinea van de Leerink note die ik hierboven plaatste:
    
    Uitgebreide preklinische gegevens rechtvaardigen de vooruitgang van QURE's kandidaat voor gentherapie met HD.     Is het laatste bericht het vervolg op de berichtgeving van november 2017?
    Gaan we nu echt nieuws over de beloofde nieuwe toepassingen horen?
    Wat is jullie inschatting op het persbericht van vandaag? Een opwarmertje? Of herhaling van oud nieuws?
    

    Ik ga maar van het positieve uit: toen was het een opwarmertje en nu heeft men de toolkit klaar voor gebruik. Ofwel: gereed voor toepassing in programma's waarmee QURE denkt te winnen.
    
    Een mogelijke strategie die men nu achter de schermen toepast, alvorens de belofte 'Expansion of the Company's early-stage research pipeline' daadwerkelijk in te lossen, is eerdere pre-klinische studies herhalen die eerder niet het beoogde therapeutische effect behaalden maar nu mogelijk met de nieuwe vectoren, promoters en methoden wel. Een voorbeeld is het eerdere AIP programma (acute interstitial pneumonia).
    
    Het zal mij dan ook niks verbazen als het échte nieuws pas in 4Q18 of 1Q19 volgt.
