   The Southwest Foundation for Biomedical Research (SFBR) has received a contract from the Dutch pharmaceutical firm Crucell to test a vaccine against Ebola and Marburg viruses, which are potential bioterror weapons. The initial contract is for $456,216 with additional potential subcontracts to be signed worth a further $2.2 million.
   
   SRBR’s Department of Virology and Immunology chaired by Jean Patterson, Ph.D., will test the immunogenicity and efficacy of a multivalent vaccine against five different strains of the viruses in animal models. SFBR’s high-level biocontainment facilities will be used to study the vaccines during the year 2010. The work is part of a $30 million primary contract awarded to Crucell by the National Institute of Allergy and Infectious Diseases of the National Institutes of Health.
   
   Crucell’s vaccine is based on inserting genetic material from the disease-causing virus or parasite into a vehicle called a vector, which then delivers the immunogenic material directly to the immune system.
   
   The Ebola and Marburg viruses are capable of causing hemorrhagic fever, a severe, often-fatal disease in humans characterized by high fever and massive internal bleeding causing death in 50 percent to 80 percent of all cases. Ebola and Marburg outbreaks occur periodically in tropical Africa, affecting both human and great ape populations. Since the Ebola virus was first recognized, approximately 2,200 cases including over 1,500 deaths have been reported. To date over 440 cases of Marburg have been reported with approximately 360 fatalities. Ebola and Marburg usually appear in sporadic outbreaks, and spread within a health-care setting.
   
   Because of the high disease-related mortality rates and lack of any vaccine or therapy, the Ebola and Marburg viruses are on the U.S. Centers for Disease Control and Prevention ‘category A’ list of bioterror agents, together with smallpox and anthrax.

   Los van beschouwingen over Crucell PR, is de inhoud natuurlijk belangrijker.
   
   Dit is vervolg op het Crucell's Ebola/Marburg vervolg contract uit October 2008 (zie hieronder), waarin Crucell's eerder werk voor 'n Ebola wordt 'overgedaan', nu met zeldzame Adenovirus vectoren ipv de initieel gebruikte Ad5.
   
   Eind 2008 contract, in 2009 ontwikkeling van de nieuwe Ad-vectoren (Ad35 bv), en in 2010 dus animal testing. Hopelijk in 2011 weer een nieuwe phase I clinical trial.
   
   Vooralsnog goed op schema, maar we staan uiteraard pas aan het begin.

   Leiden, The Netherlands (October 3, 2008) - Dutch biopharma company Crucell N.V. (Euronext, Nasdaq: CRXL; Swiss Exchange: CRX) today announced that it received a National Institutes of Allergy and Infectious Diseases (NIAID)/National Institutes of Health (NIH) contract aimed at advancing the development of a multivalent filovirus vaccine that includes both Ebola and Marburg viruses. The contract provides funding of up to $30 million, with additional options that may be triggered at the discretion of the NIH worth a further $40 million.
   
   "We are grateful to the NIAID/NIH for this award, which makes use of Crucell's proprietary AdVac® technology" said Dr. Jaap Goudsmit, Crucell's Chief Scientific Officer. "This award recognizes the scientific basis for using rare adenovirus serotypes to develop vaccines." Dr. Goudsmit added, "The contract builds upon earlier work Crucell has performed with the Vaccine Research Center at NIH, and brings us a step closer to being able to provide effective countermeasures against a highly lethal infectious disease."
   
   Crucell will be the primary contractor with additional services being supplied by the Southwest Foundation for Biomedical Research, located in San Antonio, Texas and Quintiles Guys Drug Research Unit, located in London.

   Toevoeging (in principe overbodig, maar je weet het nooit):
   Het overdoen van al het eerdere werk aan Ebola vaccin is NIET de schuld van Brus, maar gebeurt op de wens van NIAID die het zekere voor het onzekere wil nemen bij de ontwikkeling van zo'n vaccin, waarschijnlijk na het Merck Ad5 Aids drama.
   Crucell management en R&D hebben zelf heel duidelijk al eerder afstand genomen van Ad5 en zich op andere Ad-vectoren gegooid, in samenwerking met Dan Barouch. Hierdoor zijn ze het nieuwe contract met NIAID niet kwijtgeraakt, maar hebben het vernieuwd.
   
   Strategisch juist dus, om op tijd het beleid aan de veranderende wetenschappelijke realiteit aan te passen, teneinde commercieel te overleven. Goed gedaan van specifiek Brus en Goudsmit (dus een pluim voor beide heren ;-), Specifiek voor allen die hier door de negativiteitsbomen het nuance-bos niet meer zien (of nix meer zien ;-)

   In termen van R&D proces noemt men zoiets terugkoppeling tengevolge van voortschrijdend inzicht... (voor zij die voorstander zijn van snelle ontwikkeling: snelheid is ondergeschikt aan haalbaarheid en uitzicht op succes van de doelstelling). Daarbij aanstippend dat Crucell als motor is blijven functioneren in het uitzetten van de krijtlijnen van het project.

   Als long Crucell-belegger hoop ik van harte dat ze dat hernieuwde traject dan wel ASAP doorlopen. In hoeverre er veel keus was voor de opdrachtverleners kan ik niet beoordelen en dus ook niet de mate van verdienste van Goudsmit (en Brus?).

   Ik weet wel dat het van een zekere durf/visie getuigt, terwijl je bedrijf nog bezig is met een Ad5-based vaccin trial, uitgebreid de nadelen van Ad5-based vaccins te bespreken (en de oplossingen, te weten zeldzame Ad vectoren, waaraan je bedrijf ook werkt), zoals Goudsmit jaren terug deed.

   “I think it's promising,” said Jean Patterson, chairwoman of virology and immunology at Southwest Foundation. “The early data showed that it was extremely promising in rodents, and that's why they needed to come to us.”
   
   Last year, Patterson and scientists from Emory University published results from a study of a different Ebola vaccine. That vaccine, engineered from bocavirus, prevented Ebola infection in mice.
   
   Patterson said the odds are good that an effective vaccine will become available.
   
   I think if we just had the resources before this, we would have had an Ebola vaccine,” she said. “There's just been no market. The virus is extremely stable. And unlike AIDS or respiratory syncytial virus, you can develop an immune response that's lifelong.”
