   DSM and Crucell Announce Record Achievement in PER.C6® Technology
   
   Heerlen/Leiden, The Netherlands, 16 June 2008 - DSM Biologics and Crucell N.V. (Euronext, NASDAQ: CRXL; Swiss Exchange: CRX) today announce another breakthrough in the production of IgG antibodies using PER.C6® technology. By employing the PER.C6® human cell line and proprietary XD(TM) technology, a record yield of over 27 grams per liter has been achieved. In March 2008 a yield of 15 grams per liter was reported.
   
   This milestone is the new manufacturing paradigm for mammalian cell culture to produce protein products effectively, where the industry has struggled to date with low yields and unstable platforms. This record surpasses all other production systems including those previously set by PER.C6® technology itself.
   
   The use of PER.C6® technology will clearly impact cost of goods for marketed products and reduce the risk of premature high capital expenditures. Higher yields mean smaller footprints for facilities, reduced risk in drug development, higher utilization of facilities and better chance of success. Drugs that would otherwise not be brought to market because they are too expensive to produce, may now find a vehicle for commercialization. More so, the glycosylation of monoclonal antibodies and proteins, produced using PER.C6® technology, is fully human.
   
   Scientists at PERCIVIA PER.C6® Development Center, a joint venture between DSM and Crucell, have shown that they can reproducibly manufacture a record amount of product with over 95% viability and unchanged product quality. This accomplishment was achieved using standard bioreactor equipment and readily available cell culture medium. The result of this effort provides the entire biotechnology industry with a reliable production platform to economically manufacture large amounts of therapeutic proteins with ease.

   Als je trouwens luistert naar Brus aan het einde van zijn betoog dan blijkt dat ze met motivatie van de PER.C6 cellen geen probleem hebben. Het zijn werklustige celletjes met een hoge productiviteit die plezier hebben in hun werk.

    Door de menselijke cellijn PER.C6 te gebruiken, in combinatie met de eigen XD-technologie, werd een opbrengst van 27 gram lgG-antilichamen per liter behaald. In maart 2008 meldden DSM en Crucell nog een maximum van 15 gram per liter. De bedrijven stellen dat de sterke verbetering een nieuw paradigma vormt voor de productie van proteinen uit zoogdiercellen.
    
    Percivia is een joint venture van DSM Biologics en Crucell, die een jaar geleden werd opgericht om de door Crucell ontwikkelde PER.C6-technologie samen verder te ontwikkelen. De technologie wordt uitgelicentieerd aan bedrijven, die daarmee de productie van monoklonale antistoffen en recombinante eiwitten willen vergroten.
    
    Door de hogere productiviteit zouden de productiekosten voor deze stoffen aanzienlijk omlaag kunnen, waardoor bepaalde therapeutische behandelingen betaalbaarder worden voor de patient, stelt DSM.
    
    DSM Biologics is een onderdeel van DSM Pharmaceutical Products, dat weer een onderdeel is van chemieconcern Koninklijke DSM nv.

    The two Dutch firms have created a technology, called PER.C6, to produce biopharmaceutical materials such as drugs and vaccines by using human cell lines instead of a chemical process.
    
    The companies said they had boosted fermentation yields in human cells to a record level of over 27 grams per litre compared with 15 grams per litre in March. Higher yields enable smaller and cheaper production units.
    
    "The use of PER.C6 technology will clearly impact cost of goods for marketed products and reduce the risk of premature high capital expenditures," the companies said.
    
    The current global market for proteins and monoclonal antibodies, which the companies are targeting, is worth $50 million to $60 billion and is estimated to grow to $200 billion by 2020. DSM has said the joint venture had sales opportunities of more than $200 million annually.
    
    Crucell is the only company that has created a human cell line on this scale. The quality of human cells as a host for drugs and vaccines is much higher, as they are more easily accepted by the human body.
    
    Most other cell lines are produced in hamster cells and have fermentation yields of between 3 and 5 grams per litre.

    Als DSM dat ECHT gelooft dan zijn ze knettergek wanneer ze nu niet als de hazewinden toeslaan en Crucell inlijven! De DSM-koers helpt ook nog leuk mee (om van die van Crucell maar niet te spreken!).

    En ING maar denken dat de Tovenaarsleerling weer bezig is geweest met de upgrade van 15/20 naar 27.
    
    Met dit laatste cijfer behoort het aandeel gewaardeerd te worden ipv de 11 range.

    Met excuses aan de doorgewinterde, door mij zeer gewaardeerde, Crucell-kenners die dit DSM-geneuzel behoorlijk déja-vu zullen vinden:
    
    Een overname door DSM lijkt nu echt een reële optie te worden (heb zelf bij DSM gewerkt in de tijd dat er alleen nog research-samenwerking met Gist-Brocades was en ik zie duidelijke parallellen t.a.v. de timing/argumenten voor een overname). Ook de lijntjes met de Jong zijn niet onbelangrijk in dit verband! Die zal heus wel inzicht kunnen geven aan de DSM-RvB in hoeverre Crucell als productie-bedrijf op het niveau van DSM getild kan worden. De gedroomde Crucell-businessunit manager is er al!
    
    Wat zouden jullie op dit moment een reële overname prijs vinden?

    Het volume is niet zo groot. Er gebeurt aan de vraagzijde opmerkelijk weinig op basis van dit bericht. Wellicht brengen aanvullende persberichten en bijgestelde analistenvisies daar op niet al te lange termijn verandering in. Zeker wanneer het financiële plaatje dankzij QV (veel?) beter gaat uitvallen dan de analistenverwachtingen en de in ieder geval bescheiden gehouden verwachtingen door het management. Eindelijk eens "over-performen" op alle fronten?

    Geldt hogere productie ook bij de aanmaak van vaccines ?
    
    Te denken dan aan Sanofi-Aventis deals flu-vaccine (20K reactor kan kleiner ?) en Rabies deal.
    
    Of moet Sanofi-Aventis bijkomende technologie kopen om naar hogere productie te gaan ?
    
    Rabies anti-bodie-contract met Sanofi zit imo goed. Immers anti-bodies worden aangemaakt door DSM en door Crucell aan Sanofi geleverd voor toevoeging aan het vaccine.
    
    Nu nog het rabies vaccine op PER.C6 laten groeien !
    
    (Ik heb maar weer wat Crucelletjes op het schap gelegd voor na 2010... )

    Misschien een domme reactie van een biotechnologische leek.
    
    Maar ik verwacht dat het bij antibodies om veel grotere productievolumes bij lagere marges gaat (ook voedingsadditieven?). Dus daar zou deze doorbraak marge-technisch dan veel belangrijker zijn en zoals DSM aangeeft sub-marginale opties opeens wel haalbaar maken.
