    AMT Provides Business Update for the First Quarter 2010

    Amsterdam Molecular Therapeutics (EuroNext Amsterdam: AMT), a leader in the field of human gene therapy, today provides its non-audited business update in compliance with the EU transparency directive. This report summarizes material events and AMT's financial position for the first quarter of 2010.

    Q1 2010 Highlights

    - EMA starts formal review of Glybera(R) dossier
    - Dossier validated by EMA on January 20, 2010
    - Day 120 questions from EMA expected second half of May 2010
    - Innovation Credit received from Dutch Government for Duchenne Muscular Dystrophy
    - Up to EUR 4 million funding, repayable only on successful commercialization
    - Supports 35% of development costs through to 2013
    - Start of Phase I/II Gene Therapy Clinical Trial for Hemophilia B
    - First patient dosed

    Business Update

    AMT's cash position* on March 31, 2010 amounted to EUR17.7 million compared to EUR22.6 million on December 31, 2009. The cash outflow in the first quarter of 2010, amounting to EUR4.9 million mainly represented operational cash flow. AMT employed 85 persons as of March 31, 2010. Total expenses in the first quarter of 2010 were EUR5.1 million compared to EUR4.9 million in the same period last year.

    * The Company's cash position is composed of cash and cash equivalents.

    Material events after March 31, 2010

    On May 6, 2010 the Company announced that Professor Sander van Deventer joined the Supervisory Board. At the same time, Alexander Ribbink retired from the Supervisory Board and Piers Morgan (CFO) was appointed to the Management Board.

    Uiteindelijk via Google moeten zoeken om het officiele persbericht te kunnen vinden. Op de website van AMT is hierover niets te vinden.

    Voor de mensen die de website niet kunnen raadplegen:
    Op de startpagina van de website staat een soort Flash element/filmpje waarin je een DNA Helix ziet met het woord CARE, waarin de A langzaam veranderd in een U, waardoor het woord CURE zichtbaar wordt. Dit dus volgens de slogan van AMT: From Care to Cure.
    Internet Explorer heeft dus klaarblijkelijk moeite met dit Flash element/filmpje.

    Gene Therapy Vector Successfully Delivers shRNA into Liver Cells

    Amsterdam Molecular Therapeutics (Euronext: AMT) a world leader in gene therapy, today reported positive preclinical data from a study using an AAV gene therapy product to lower cholesterol. The data show that a single dose of the gene therapy carrying a short hairpin RNA to silence Apolipoprotein B100 (ApoB100), resulted in a reduction of serum cholesterol of approximately 80%, without any signs of toxicity. These data validate AMT’s technology platform as a powerful tool for successful gene silencing within target cells. The data were presented by Annemart Koorneef, scientist at AMT, yesterday during the 13th Annual American Society of Gene and Cell Therapy (ASGCT) Meeting in Washington, DC.

    “With just a single dose of our cholesterol targeting gene therapy, a long-lasting, significant reduction of serum cholesterol is achieved. This preliminary study suggest that AMT’s technology may have overcome one of the major problems of shRNA therapies, namely efficient and non-toxic intracellular delivery,” noted Jörn Aldag, CEO of Amserdam Molecular Therapeutics.

    In the study, AMT used its proprietary AAV-based platform to efficiently deliver a shRNA that silences both human and mouse Apolipoprotein B100 (ApoB100). A single intravenous administration caused prolonged ApoB100 gene silencing that was sequence-specific and not associated with liver toxicity, oversaturation of the cellular miRNA machinery or induction of immune responses. In addition, it was shown that the shRNA constructs against ApoB100 specifically and efficiently silence human ApoB100 ex vivo.

    ApoB100 is the structural protein of Low Density Lipoprotein (LDL) particles that carry cholesterol. Silencing of ApoB100 with shRNAs results in a reduction of LDL-cholesterol and has the potential to be used to treat hypercholesterolaemia and cardiovascular disease.
